Summary

Model-informed approaches integrate QSP, PBPK, population PK/PD, joint models and model-based meta-analysis to strengthen strategic decisions in oncology drug development, from preclinical translation to phase III trial design. These methods enable quantitative interpretation of heterogeneous preclinical data, justification of first-in-human and phase 2 doses, and more precise prediction of late clinical endpoints such as overall survival. In oncology, where productivity and success rates remain low, integrated efficacy and safety modelling provides a robust basis for optimised dosing strategies and more informative go/no-go decisions.
