The current work represents the first-in class multilevel mechanistic model of branched-chain amino acids (BCAA) catabolism, verified using historical as well as de novo generated nonclinical data. The developed platform enables not only optimization of dosing regimens for BCAA-targeted therapies but can be also used to inform design of further experiments. Mechanistic nature of the model allows evaluation of interactions between enzymes and substrates and therefore identification of factors affecting treatment outcomes.
